GERBU Adjuvants

Antigen immunotherapyIn 1991 Dr. Nikolaus Grubhofer started research to make his vision come true. He decided to develop a better adjuvant, which meant: match Freund’s adjuvant as the yardstick of efficiency, make it perfectly biotolerable, remove the emulsion handling problem, maintain a reasonable price level and get it patented and licensed. Many formulations and data has become available as the result of the 50 years of scientific work following Freund.

General Information

Product inserts supplied with the product give full details of the injection protocol. All GERBU Adjuvants are powerful potentiators of cellular and humoral immune response with long term protecting effect. Very mild and simple to handle, just mix with antigen and inject. All components are nontoxic and fully biocompatible. Requires frequent boosting to achieve a high-titered response.

Active Ingredients:

The effect of GERBU adjuvant relies on the synergy between the glycopeptide GMDP (N-Acetyl-glucosaminyl-(β1,4)-N-acetylmuramyl-l-alanyl-d-isoglutamine), originally isolated from Lactobacillus bulgaricus, and solid microfiltrable particles of biodegradable lipids consisting of paraffines. Also important are the biocompatible emulsifiers and the synergistic and stabilizing environment.

Suggestions for Use:

Mixing with antigen-solution: Volume ratio of 1:1 to the antigen solution usual but not mandatory.

Withdrawal of the adjuvant: By use of a sterile plastic syringe, the required volume is transferred into a plastic vial. Mixing with the antigen solution is done by drawing up into a syringe several times. No emulsion to prepare – just mix with antigen.

Dilution: A volume-ratio of 1:1 to the antigen-solution is usual. Nevertheless it is possible to adjust the antigen-volume to the concentration of the antigen, as long as the recommended antigenvolume is retained and the maximum volume at the injection site is not exceeded. Diluent: Water at best, possibly buffered with HEPES, MOPS or glycine. Avoid buffers with polyvalent anions like phosphate or citrate-they might provoke flocculation of the positive charged particles. A pH value between 5 and 6 has been proved to be more favorable than strictly neutral. Ready-made mixtures should be stored frozen.

Application: Subcutaneous application is recommended. Intramuscular injection usually does not work better, but is rather more harmful the animals. e.g. chicken might discontinue laying eggs after i.m. injections into the breast muscle

Storage: Storage at 4-8°C is recommended. At this temperature, the adjuvant becomes viscous. Under these conditions, the particle suspension is highly stable. Prior to the use of the adjuvant, the solution must be at room temperature.

The GERBU Adjuvant for monoclonal antibodies has been shown to give high yields of IgG and good fusion – yields especially in mice. Adjuvant M is supplied in tubes of 0.1 ml, permitting processing of even the smallest amounts of antigen without loss. Additionally available are vials of 1 ml in order to withdraw the required aliquot volume as well as bulk sizes at favorable prices for commercial users.
